I don't really get what Sakurai meant by that.

I think it means this:

Once you defeat a character, it joins your "team". Like, once you save Zelda/Peach, you have either one of them on your team. And then, you can switch out with them after a level. That help?

But do you think that at the end of each level you'd be able to switch between anyone on your team at the end of a level.

We don't know. Members could mean number of characters on team, but there's only two in that photo. Another option is the amount of lives you have. It isn't discernible at this point.
